Characterization of genetic variation among accessions of Aegilops tauschii

The application of AFLPs, and SSRs to examine genetic relationships in the 54 accessions of Aegilops tauschii from 5 countries was investigated. A total of 256 (234 polymorphic) and 224 (219 polymorphic) bands were detected using 17 primer combinations of AFLP and 24 well distributed, mapped SSR markers, respectively. The UPGMA cluster plots separated the 54 accessions into three major groups based on their AFLP fragment similarities, and into five major groups based on their SSR genotypic similarities. Correlation between both genetics similarity matrices was poor. Both of molecular techniques did not group accessions strictly according to their geographical origins. These different marker systems should provide different levels of information which is important in management of germplasm resources.
